| Sumario: | The auxin system for inducible protein degradation is a powerful tool for investigating protein function in budding yeast. Variants of all system components are available, but which ones should be combined for optimal results is unclear. - - The authors systematically evaluated various system components and found that the degrons mIAA7 or AID* together with adamantyl-auxin and the degron receptor OsTIR1(F74G) allow particularly rapid and extensive protein depletion, with little basal degradation. Moreover, they demonstrated that the system can replace chemical inhibitors and temperature-sensitive alleles. - - This study clarifies how the auxin system should best be used and will aid its effective application. |
